Completed an oil change last week and added oil until the dipstick read full. Even days later prior to really driving it the oil stick still read full. Today I took it for a 30 minute drive since I will be leaving on a 12 hour drive next weekend and checked the oil when I got home. Now it reads above full.
On my other trucks there is a hot and cold side to the dip stick with hot being larger as the oil gets hot. Since this one does not have a hot and cold side does this mean that I am the dipstick here?
Should I drain some of the oil out?

If it is just a little over, I'd run it. If it is filling up and looks like it is going to overflow, you probably have an injector O ring leaking fuel in to the oil. Assuming that it is the engine oil you are talking about.

Yes, I think you should drain some back out to get it down to full. They hold 15 quarts with a filter change. These trucks are bad about hanging oil in the heads and lines so it takes a while for it to drain back to the pan. Next time add 15 quarts ( I put 1 quart in the filter before install so I would only add 14 quarts) and them let it drain back down for a few minutes before you check it.

I think that it is close to 16 qts and it is about 1/2 in over full. I am going to drain off a quart or so. Do you think that my 20 mile drive with that extra quart hurt anything?

You didn't hurt any thing. Next time just put in 15 qts. Oil expands when it gets hot. Allow for that.

I always run mine at the bottom crosshatch area of the oil stick, the lower level decreases the chances of aireation of the oil which it turn reduces fuel economy and power levels.
