You can either loosen each injector line one at a time while the engine is running to listen for a drop in RPM, or you can use a voltmeter set to millivolts to probe the GP spades (with the GP leads disconnected) to see if they're putting out any voltage. It would be in the 0.5-5.0 millivolt range usually, and all GPs should read relatively even. If you have a reading that is way off, you can swap that glow plug with one from one of the 'normal' cylinders and retest it. If the abnormal reading moves with the GP, you have a bad GP, but if it stays with the cylinder then there is something wrong with the injector in that hole. This test is done once the engine is fully warmed (so it'll start w/o GP harness connected).

A multimeter with a millivolt scale will do fine. There's no specific RPM you're looking for, you're just listening for a change. The RPM should drop when you loosen the line going to a good injector.

The best way to know if your injectors are firing evenly, is to yank them out have have them pop tested. You can make you own tester or take them to a shop and they usually will just do it free or for a very small amount.
I found a VW forum that someone has a picture and a whole writeup using a cheap bottle jack and lots of other odds and ends to make a home made tester.
