Is this the Heater Core case/cover, or the blower cover? This is located in the engine bay right above the heater inlet hose?? Can i use some type of fiberglass to close this hole?

Thats what I would do. Looks like the coil is exposed. Try to leave an air gap when glassing up, so air can still flow by, internally.
