Is there any difference between the 4R100 in an E350 and and F350? Also does the motor matter if it is going to be rebuilt? Basically Im looking for a used tranny to tear down and rebuild myself with upgraded parts, and found one out of an E350 V10. Also does it matter 2WD or 4WD?

A trans from a V10 will bolt to a V10 or a 5.4L. It will not bolt to a diesel. A 4x4 trans will need the output shaft and extension housing changed to go into a 4x2 truck.
E and F series 4x2 transmissions are the same as long as the engines are the same.
